Designing a Multi-Epitope Subunit Vaccine against VP1 Major Coat Protein of JC Polyomavirus
نویسندگان
چکیده
The JC polyomavirus virus (JCPyV) affects more than 80% of the human population in their early life stage. It mainly immunocompromised individuals where replication oligodendrocytes and astrocytes may lead to fatal progressive multifocal encephalopathy (PML). Virus protein 1 (VP1) is one major structural proteins viral capsid, responsible for keeping alive gastrointestinal urinary tracts. VP1 often targeted antiviral drug vaccine development. Similarly, this study implied immune-informatics molecular modeling methods design a multi-epitope subunit targeting JCPyV. epitopic sequences, which are highly conserved, were used build vaccine. This designed includes two adjuvants, five HTL epitopes, CTL BCL epitopes stimulate cellular, humoral, innate immune responses against Furthermore, dynamics simulation (100 ns) studies examine interaction stability with TLR4. Trajectory analysis showed that TLR4 receptor form stable complex. Overall, contribute path development

Hydroxyproline in the major capsid protein VP1 of polyomavirus.
Amino acid analysis of [3H]proline-labeled polyomavirus major capsid protein VP1 by two-dimensional paper chromatography of the acid-hydrolyzed protein revealed the presence of 3H-labeled hydroxyproline. Addition of the proline analog L-azetidine-2-carboxylic acid to infected mouse kidney cell cultures prevented or greatly reduced hydroxylation of proline in VP1. Immunofluorescence analysis per...
متن کاملImmunoinformatics Design of a Multi-epitope-based Vaccine Against Colorectal Cancer
Background: Bioinformatic approaches for designing vaccines have become a promising alternative to conventional methods. We herein designed a multi-epitope-based vaccine against colorectal cancer (CRC). Methods: Used peptides in the CRC vaccines were retrieved from databases of PubMed, Web of Science, Google Scholar, and Clinical trials. The adjuvants of Mycobacterial heparin-binding hemagglut...
متن کاملDesign of a Multi-epitope Peptide Vaccine against SARS-CoV-2 based on Immunoinformatics Data
Background and purpose: In 2019, the world has witnessed the emergence of a virus that caused acute respiratory distress syndrome in human with high mortality rates (approximately 3.7%). So far, no effective treatment has been proven against COVID-19. This study aimed at designing a multi-epitope vaccine combining several T-cell and B-cell epitopes of the SARS-CoV-2. Materials and methods: Bas...
متن کاملDesign of a Novel Recombinant Multi-Epitope Vaccine against Triple-Negative Breast Cancer
Background: Triple-negative breast cancer (TNBC) is determined by the absence of ERBB2, estrogen and progesterone receptors’ expression. Cancer vaccines, as the novel immunotherapy strategies, have emerged as promising tools for treating the advanced stage of TNBC. The aim of this study was to evaluate Carcinoembryonic antigen (CEA), Metadherin (MTDH), and Mucin 1 (MUC-1) proteins as vaccine ...
